A STUDY ON THE RELATIONSHIP OF THIN SHEET OF DOUGH MICROBIOLOGICAL QUALITY BETWEEN QUALITY OF RAW MATERIAL AND SOME HYGIENE PARAMETERS

The thin sheet of dough has been recently produced at home and small businesses, today it is converted into an
industrial product. The present study, the relationship ofthe microbiological properties between the raw materials
(flour and water) and hygiene parameters (surface, equipments, personnel hands and air) with thin sheet of dough (end-product) were firstly investigated in thin sheet of dough production facilities (A, B and C) in Turkey. Thin sheet of
dough, the highest mean Total mesophilic anaerobic bacteria (TMAB), coliform bacteria, Staphylococcus spp.,
Staphylococcus aureus and mould counts were 4.4x10

cfu/g, respectively. In wheat flour samples, the highest mean TMAB, coliform bacteria, Staphylococcus spp.,

cfu/g, respectively. Moreover, significant differences were found about TMAB, Staphylococcus spp. and
mould counts between wheat flour and thin sheet of dough samples (P<0.05). The highest mean TMAB count was

cfu/g in water samples and the coliform bacteria and Esherichia coliwere not found in water
samples. In samples taken from staffs in food production area, the highest TMAB and coliform bacteria counts were
found in ‘A’ company. Coliform bacteria on the examined personnel hands were found 1 cfu/cm

in 13 (36.1%)
samples, 5 cfu/cm

in 20 (55.6%) samples, 45 cfu/cm
2
in 3 (8.3%) samples and S. aureus counts were found 1 cfu/cm

in 29 (80.6%) samples, 5 cfu/cm

in 7 (19.4%) samples. The highest mean TMAB and coliform bacteria counts were
detected in end-product samples taken from ‘B’ company and these microorganisms counts were found also high in
wheat flour, surface, equipment and personnel samples taken from ‘B’ company. Similarly, the highest mean
Staphylococcus spp., S. aureusand mould counts were found in ‘A’ company and these results was correlated with
wheat flour, personnel and air samples counts result. Accordingly, there is necessity to improve the process and
sanitary conditions in manufacturing with to standardize of the using raw materials for the qualified thin sheet of
dough production.

Yufka, kısa bir süre öncesine kadar ev ve küçük işletmelerde üretilmekte iken, günümüzde modern işletmelerde üretilen endüstriyel bir gıda ürünü olarak karşımıza çıkmaktadır. Bu çalışma ile Türkiye’de ilk kez yufka üretim tesislerinde (A, B ve C) hammadde (un ve su) ve hijyen parametrelerinin (yüzey, ekipman, personel ve hava) mikrobiyolojik özellikleri ile yufkanın (son ürün) mikrobiyolojik özellikleri arasındaki ilişki araştırılmıştır. Yufka örneklerinde (n: 9) en yüksek ortalama Toplam Mezofilik Aerob Bakteri, koliform bakteri, stafilokok, Staphylococcus aureus ve küf sayıları sırası ile 4,4x104 kob/g, 1,7x101 kob/g, 9,3x102 kob/g 1,7x102 kob/g, 6,9x103 kob/g olarak belirlenmiştir. Un örneklerinde ise (n: 9), ortalama TMAB sayısı 1,5x104 kob/g, ortalama koliform bakteri 4,4x101 kob/g, ortalama stafilokok 9,3x102 kob/g, ortalama S. aureus sayısı 1,0x102 kob/g ile ortalama küf sayısı 6,8x103 kob/g olarak saptanmıştır. Çalışmada her üç işletmeden elde edilen un ve yufka örneklerinde tespit edilen mikrobiyolojik sonuçlar arasındaki farklılığın TMAB, stafilokok, küf sayıları açısından önemli olduğu bulunmuştur (P<0,05). Su örneklerinde en yüksek ortalama TMAB sayısı 1,7x101 kob/g iken, koliform ve E. coli saptanmamıştır. Yüzey ve ekipman örneklerinde en yüksek TMAB ve koliform sayısı A işletmesinden tespit edilmiştir. Personel el örneklerinde koliform bakteri ve S. aureus sayısı sırasıyla 13 (% 36,1) örnekte 1 kob/cm2, 20 (% 55,6) örnekte 5 kob/cm2, 3 (% 8,3) örnekte 45 kob/cm2; 29 (% 80,6) örnekte 1 kob/cm2, 7 (% 19,4) örnekte ise 5 kob/cm2 olarak bulunmuştur. Hava örneklerinde en yüksek ortalama TMAB ve küf sayısı 1.6x102 kob/petri plak ve 8,5 x101 kob/petri plak olarak belirlenmiştir. Buna göre son üründe en yüksek ortalama TMAB ile koliform bakteri sayısı B işletmesinde saptanmış ve aynı işletmeden alınan un, yüzey, ekipman ve personel örneklerinde de söz konusu mikroorganizma sayılarının yüksek olduğu belirlenmiştir. Benzer şekilde A işletmesinden temin edilen yufka örneklerinde yüksek düzeyde saptanan stafilokok, S. aureus ile küf sayısının, işletmeden elde edilen un, personel ve hava örneklerini sonuçları ile ilgili olduğu gözlenmiştir. Buna göre kaliteli yufka üretimi için, proses ve sanitasyon şartlarının geliştirilmesi ile kullanılan hammaddelerde standardizasyonun sağlanması gerekmektedir.
