Leukoplakia which is defined as white patch or plaque of oral mucosa that cannot be characterized clinically as any other disease, is the most common lesion of oral mucosa that has the premalignant and malignant potential. The effect of smoking is well known in the etiology of leukoplakia, however the role of many agents is still controversial. In recent years, the role of viruses with the special reference to Human papillomavirusfHPV} has been discussed. The aim of present study is to evaluate the presence of HPV in oral leukoplakia and to discuss its role in etiopathogenesis. In (he present study, the routine hematoxylinc&eosine stained slides of a total of 20 biopsies of cases with oral leukoplakia lesions were histologically reviewed for the conformation of diagnosis of leukoplakia. Twenty paraffin embedded blocks containing representative histological specimens of the lesions were selected and stained immunohistochemically for pan HPV and HPV 16/18 E6 protein. The results of histopathological assessment showed that there were 7 cases of dysplasia, 11 cases of hyperkeratosis, 1 case of verrucous hyperplasia and 1 case of candidal leukoplakia. Thirteen of 20 cases demonstrated positive immunohistochemical staining for pan HPV, HPV 16/18 E6 pratem was positive in 5 of 20 cases. Oral mukozanı n bilinen diğer lezyonları ile karakterize edilemeyen beyaz lezyonlan olarak tanımlanan lökoplaki, oral mukozanın en yaygın premalign ve malign potansiyeli alan lejyonudur. Lökoplaki etyolojisinde iyi tanımlanmış bir faktör olan sigaranın dışında bir çok faktör hala tartışılmakladır. Son yıllarda viral ajanların, özellikle Humarı papülomavirus'un (HPV) lökoplaki etyopatoge-rtezine ve malign transformasyona katkısı araştırılmaktadır. Bu çalışmada oral lökoplakilerde imrniintıistokimyasa.1 olarak HPV varlığını ve virüsün etyopatogeneze etkisini araştırmak amaçlanmıştır. Çalışmamızda toplam 20 lökoplaki ön tanısı alını; vakanın histopatolojik tanılan değerlendirilmiş ve bu vakalara ait parafin bloklardan alman kesitlerde pan HPV ve malign transformasyonda etkisi olan HPV 16718 E6 protein immünbistokımyasal olarak boyanmıştır. Histopato lojik değerlendirme sonucu 7 vaka displayj tanısı alırken, 11 vaka hiperkeratotik mukoza, 1 vaka verrüköz hiperplaTİ, CAlbicans varlığı saptanan 1 vaka ise kandidal lökoplaki tanısı almıştır. Yapslan immünhistokimyasal boyamalar sonucunda 13 vakada pau HPV pozitifliği, 5 vakada ise HPV 16/18 E6 protein pozitifliği .saptanmıştır. Vakaların 4'ü ise hem pan HFV hem de HPV 16/18 pozitifliği göstermiştir.
