Objectives: Diabetes mellitus is a systemic disorder. Vascular complications of diabetes are not rare. In this study, we aimed to assess the effect of long-term diabetes (14 weeks) on vascular reactivity in ischemia-reperfusion model. Methods: Twelve Sprague-Dawley male rats divided into two groups as diabetes and control. 2 rings with endothelium, 2 rings without endothelium gained from thoracic aorta of every rat. Vascular response to phenilephrine, acetylcholine and nitroprusside was assessed. Results: There was no statistically significance for sodium-nitroprusside relaxation response between diabetes and control groups before ischemia. However, after ischemia there was minimal significance (p=0.049). No statistically significances were observed for other molecules between two groups (p>0.005). Conclusion: We conclude that in order to understand the effects of diabetes mellitus on the endothelial response in ischemia-reperfusion model new studies should be planned on diabetic models with longer duration.

Amaç: Diyabetes mellitus sistemik bir hastalıktır. Bu hastalıktaki vasküler komplikasyonlar hem oldukça sık, hem de mortalitenin en önemli nedenleri arasındadır. Bu çalışmada uzun süreli (14 haftalık) oluşturulan, deneysel diyabetin iskemi reperfüzyon modelinde, endotel fonksiyon bozukluğuna etkisi araştırıldı. Yöntem: Ondört haftalık diyabet oluşturulmuş grupta 6 ve kontrol grubunda 6 olmak üzere toplam 12 Sprague-Dawley cinsi erkek sıçanın torasik aortalarından endotelli ve endotelsiz olmak üzere ikişer halka alındı. Bu iskemi-reperfüzyon modelinde fenilefrin, asetilkolin ve sodyum nitroprussid ile damar halkalarının cevabına bakıldı. Bulgular: Kontrol ve Diyabetik gruplarda, iskemi öncesi maksimal sodyum-nitroprussiad gevşeme yanıtları değerlendirildiğinde, anlamlı bir fark yok iken, iskemi sonrası yanıtlarında sınırda bir farklılık bulunmuştur (p=0.049). Diğer fenilefrin kasılma ve asetilkolin gevşeme yanıtlarında ise, gruplar arasında anlamlı bir fark gözlenmedi (p>0.05). Sonuç: Diyabetin, iskemi-reperfüyon modelinde endotel cevabına etkisinin anlaşılabilmesi için yapılacak çalışmaların, daha uzun süreli oluşturulmuş diyabet modellerinde yapılması daha faydalı olabilir.
