Objective: In this study, we investigated the effects of intramusculer use of PGE1 in experimental left renal ischemia-reperfüsion injury in rats histologically and biochemically. Materials and Methods: 28 adult female Wistar rats with regular estrous cycles were randomly divided into 4 groups. Group 1 was the control group. Group 2 , ischemia was produced for 60 minutes. Group 3 , 60 minutes , ischemia was produced and than 60 minutes reperfused. Group 4; ischemia and reperfusion was produced for 60 minutes. Before fifteen minutes of reperfusion 20 mcg/kg/IM PGE1 was performed. For light microscopic investigation, renal samples put into %10 formaldehyde solutions. Glomerular and tubular injury was histologically determined with light microscopy. Results: Light microscopic investigations showed that the renal damage in the ischemia group was more than the control group. But the most extensive damage occured at ischemia- reperfusion group. In PGE1 group the damage was less than ischemia-reperfüsion group. Conclusion: In our study, the histological and biochemical analyses revealed that PGE1 was an effective antioxidan in experimental renal ischemia-reperfüsion injury in rats to reduce the damage. ©2004, Fırat Üniversitesi, Tıp Fakültesi
Abstract (Original Language): 
Amaç: Bu çalışmada, ratlarda deneysel olarak oluşturulan sol böbrek iskemi-reperfüzyon hasarında, antioksidan olarak intramüsküler PGE1 kullanımının ışık mikroskopik ve biyokimyasal etkilerini araştırmaktır. Gereç ve Yöntem: 28 adet erişkin, Wistar albino cinsi dişi rat, estrus fazında, rastgele dört gruba ayrıldı. Grup 1 kontrol, Grup 2, sol renal arter 60 dakika klemplenerek iskemi, oluşturuldu. Grup 3, bir saat iskemi-bir saat reperfüzyon uygulandı. Grup 4 bir saat iskemi uygulanıp bir saatlik reperfüzyondan 15 dakika önce 20mcg/kg/IM PGE1 verilen grup. Tüm ratlardan alınan sol böbrek doku örnekleri ışık mikroskopik inceleme için %10'luk formaldehit solüsyonuna konuldu. Işık mikroskopik inceleme glomerül yapısında ve tübüler yapıdaki bozuklukların varlığı açısından histolojik olarak değerlendirildi. Bulgular: Işık mikroskobik incelemede sol böbrekte iskemi yapılan grupta, kontrol grubuna göre anlamlı olarak hasar tesbit edildi. En şiddetli hasar ise, iskemi-reperfüzyon grubunda tesbit edildi. PGE1 kullandığımız grupta hasar, iskemi-reperfüzyon grubundan daha az idi. Sonuç: Sol böbrekte iskemi oluşturmak dokuda hasarlanmaya yol açmakta, iskemi-reperfüzyonda ise bu hasarlanma daha da artmaktadır. PGE1 kullanımı bu tasarlanmaları anlamlı olarak azaltmaktadır.
