To compare the enzymo-histochemical and electrophoretic methods to determine the contractile properties of skeletal muscle related to training background, 26 male and 8 female, totaly 34 runners volunteered for the study. Muscle samples obtained from vastus lateralis muscles of subjects by the percutaneous needle biopsy technique were analysed by myofibrillar ATPase staining (mATPase) and sodium dodecyl sulphate polyacrylamide gel electrophoresis (SDS-PAGE) methods and fibre composition of the samples were calculated. In addition, muscle strength of the knee extansor muscles of the subjects were measured at 30°/sec and 240°/sec of angular velocites. The results showed that; 1) muscle fibre composition determined by both mATPase staining and myosin heavy chain composition reflects the differences in training background, 2) both methods do not bring out any differences to state muscle contractile properties related to training background for type I fibre whereas they bring out different comments for the subgroups of type II fibers. In the light of these findings, it may be concluded that to state the muscle contractile properties related to training background, it is not adequate to determine the fibre type composition of the muscle by only enzymo-histochemical method.

İskelet kasının antrenman geçmişi ile ilişkili kontraktil özelliğini belirlemede kullanılan enzimohistokimyasal ve elektroforez yöntemlerinin karşılaştırıldığı bu çalışmaya 26'sı erkek, 8'i bayan olmak üzere 34 elit atlet gönüllü olarak katıldı. Deneklerin perkutan iğne biyopsisi tekniği ile vastus iateralis kaslarından alınan örnekleri miyofibhller ATPase (mATPase) boyaması ve sodyum duodecil sülfat poliakrilamid jel elektroforez (SDS-PAGE) yöntemi değerlendirilip kas lifi kompozisyonları hesaplandı. Ayrıca deneklerin diz extansör kaslarının 30°/san ve 240°/sn'lik açısal tuzlardaki kuvvetleri ölçüldü. Sonuçlar; 1) antrenman geçmişindeki farklılıkların mATPase boyaması ile tespit edilen kas lifi dağılımı ve ağır zincir myozin kompozisyonunun her ikisine de belirgin bir şekilde yansıdığını, 2) antrenman geçmişi ile ilişkili kan kontraktil özelliklerini ifade etmek için bu iki yöntemin tip I kas lifleri açısından farklı bir boyut getirmezken tip II kas liflerinin alt grupları için farklı yorumlar ortaya koyabildiğini gösterdi. Bu bilgiler ışığında iskelet kasının antrenman geçmişi ile ilişkili kontraktil özelliklerini ifade etmede enzimo-histokimyasal olarak kas lifi kompozisyonunu belirlemenin tek başına yeterli bir parametre olmayacağı sonucuna varıldı.
