The Effects of Moderate and Strenuous Running Exercise on Muscle and Erythrocyte Oxidant/Antioxidant Status

Background: This study was designed to investigate the effects of duration and intensity of training on oxidant status and antioxidant enzyme activities of muscle and erythrocyte. Material and methods: Twenty five male Wistar-albino rats were divided into three groups as a control (n=10), moderate (n=8) and strenuous (n=7) exercise. Moderate and strenuous exercise groups were trained for half an hour and one hour throughout thirteen weeks, respectively. The gastrocnemius muscle tissues and the erythrocytes were taken for spectrophotometeric analyses of SOD, CAT, GSHPx activities, and NO and thiobarbituric acidreactive substances (TBARS) levels after the experimental procedure ended. Statistical evaluations were done by Mann-Withney-U and Spearman rank tests. Results: SOD and GSH-Px activities in erythrocyte of two experimental groups significantly increased, whereas TBARS levels significantly decreased. Catalase activity only increased slightly in muscle tissue of moderate exercise group. There was a negative correlation between SOD activity and NO level of gastrocnemius muscle in moderate exercise group. In strenuous exercise group, we found negative correlations among SOD and GSHPx activities and NO level in muscle and erythrocyte tissues. Conclusion: Moderate exercise has great influence on antioxidant enzyme activities rather than the strenuous exercise. However, strenuous exercise plays a regulator role in balance between oxidant and antioxidant status.

Amaç: Sıçanların kas ve eritrosit antioksidan enzim aktivitesi üzerine antrenman süre ve şiddetinin etkisini göstermek için çalışma planlandı. Gereç ve yöntem: Çalışmada kullanılan yirmi beş adet erkek Wistar-albino sıçan; kontrol, orta ve yüksek şiddette antrenman yaptırılmak üzere üç gruba ayrıldı. Orta ve yüksek düzeyde antrenman yaptırılan sıçanlar on üç hafta boyunca günde yarım ve bir saatlik süre ile deney protokolüne bağlı olarak koşu bandı eğimi değiştirilerek koşturuldu. Çalışmanın sonunda eritrosit ve gastroknemius kasından alınan örneklerden superoksit dismutaz (SOD) ve glutatyon peroksidaz (GSHPx) aktiviteleri, nitrit/nitrat, tiobarbiturik asit türevi aktif maddeler (TBARS) düzeyleri ve kas katalaz (CAT) aktivitesi spektrofotometrik yöntemle ölçüldü. Gruplar arasındaki farklılığı değerlendirmek için Mann-Withney-U ve parametrelerin korelasyonu için Spearman rank testi kullanıldı. Bulgular: Her iki antrenman grubunda eritrosit SOD ve GSH-Px aktivitesinin artmasına karşılık TBARS düzeyleri azaldı. Orta şiddette antrenman yapan grubun gastroknemius kas dokusu CAT aktivitesi artış gösterdi. Orta düzeyde egzersiz yaptırılan sıçanların kas CAT ile nitrik oksit seviyeleri arasında negatif bir korelasyon tespit edildi (r=-0,743 p<0,05). Ayrıca günde yüksek düzeyde egzersiz yaptırılan sıçanların eritrosit GSH-Px ve SOD aktiviteleri ile NO düzeyleri arasında negatif korelasyon görüldü (r=-0,865 p<0,012 ve r=-0,847 p<0,016). Sonuç: Orta düzeyde egzersizin antioksidan aktivite üzerinde daha etkili olduğu, ancak; yüksek düzeyde antrenmanlı hayvanlarda oksidan/antioksidan sistemler arasındaki dengenin daha iyi düzenlendiği saptanmıştır.
