Flow-cytometric analysis of reticulated platelets in estimating thrombopoiesis. Thiazole Orange (TO), a fluorescent dye originally used for reticulocyte analysis, is characterized by a large fluorescence enhancement yield on binding to nucleic acids, particularly RNA. We app¬lied TO staining to platelets in whole blood samples from haematologically normal subjects and patients with various quantitative platelet disorders. The percentage of TO+ platelets in 58 control subjects was %3.85(1.99(±SD). In 27 thrombocytopenic patients whose bone marrovv contained normal to increased number of megakaryocytes, the percentage of TO+ platelets %16.93(12.93(±SD), was significantly elevated (p<0.0001). In contrast, the proportion of po-sitively stained platelets in 17 patients with thrombocytopenia due to various conditions with reduced marrovv megakaryocytes did not significantly differ from the controls, whereas the ab-solute counts of TO+ platelets were significantly lovvered (p(0.0001). The percentage of TO+ platelets in 15 patients with thrombocytosis was % 3.65+2.28 (+SD) and did not significantly differ from controls, but the absolute counts of TO+ platelets were significantly increased (p<0.0001). We conclude that flow cytometric analysis of reticulated platelets is a sensitive and spesific test that rapidly provides information on the thrombopoietic activity in thrombocytopenic disorders

Thiazole orange (TO) nükleik asitlere, özellikle RNA ya bağlandığında fluoresans oluşturan, aslında retikülosit sayımında kullanılmış bir boyadır. TO boyamasını hematolojik olarak normal kişilerde ve çeşitli niceliksel trombosit bozukluğu olan hastalarda tam kan örneklerinde kullandık. 58 kontrol örneğinde TO ile boyanan (TO+) trombositlerin oranı %3.85±1.99(±SS) bulundu. Kemik iliğinde megakaryosit sayısı normal veya artmış olan 27 trombositopenik hastada TO+ trombosit oranı %16.93±12.93 bulundu. Bu kontrole göre anlamlı olarak yüksekti (p<0.0001). Kemik iliğinde megakaryosit sayısının azaldığı 17 trombositopenik hastada TO+ trombosit oranı kontrolden farklı bulunmadı. Fakat mutlak TO+ trombosit sayısı anlamlı olarak düşüktü (p<0.0001). 15 trombositozlu hastada TO+ trombosit oranı %3.65±2.28 (±SS) bulundu ve bu kontrol değerlerinden farklı değildi. Yine bu hasta grupunda TO+ trombositlerin mutlak sayısı kontrol değerlerine göre anlamlı olarak yüksek bulundu (p<0.0001). Sonuç olarak retikülotrombositlerin akım sitometrisi yöntemi ile sayımı sensitif ve spesifik bir test olup, trombositopenik hastalarda trombopoetik aktiviteyi noninvazif olarak hızlı bir şekilde değerlendirmeyi sağlamaktadır.
