Biberiye ve Adaçayı Ekstraktlarının Ruminal Fermantasyon Üzerine Etkilerinin Rumen Simülasyon Tekniği (RUSITEC) ile Araştırılması

The aim of this study was to determine the effects of rosemary (Rosmarinus officinalis L.) and sage (Salvia officinalis L.) extracts on in vitro ruminal fermentation using the long-term rumen simulation technique (RUSITEC). The RUSITEC system was equipped with six fermenters, each with a capacity of 1000 ml. Each fermenter received daily 5 g barley straw and 5 g barley. The experiment lasted 12 days. After an adaptation period of 6 days, the fermenters divided into 3 groups, first two vessels received no additives (control), second two vessels received 250 mg rosemary extract daily, and third two vessels received 250 mg sage extract daily. During the experimental period rumen fluid pH and protozoa number were determined and samples were collected for the analysis of ammonia nitrogen (NH3-N), volatile fatty acids (VFA) and dry matter digestibility. Rosemary and sage extracts had no effect on ruminal pH, total-VFA, propionate and butyrate production, total protozoa number, NH 3-N concentration and dry matter digestibility. However, acetate production and acetate/propionate ratio decreased (p < 0.05) when compared with the unsupplemented control fermenters. The results of this study showed that rosemary and sage extracts exerted beneficial effects on some fermentation parameters in the RUSITEC system. If these effects were also induced when these substances are added to the rations of rumin ants, beneficial changes in the animals performance may be expected.

Bu araştırmanın amacı, uzun süreli bir inkübasyon modeli olan rumen simülasyon tekniğini (RUSITEC) kullanarak biberiye (Rosmarinus officinalis L.) ve adaçayı (Salvia officinalis L.) ekstraktlarının rumendeki mikrobiyal fermantasyon üzerine in vitro etkilerini belirlemektir. RUSITEC sisteminde hacimleri 1000 ml olan 6 fermenter kullanıldı. Her bir fermenterde günlük olarak 5 g arpa samanı ve 5 g arpadan oluşan bir deneme yemi karışımı inkübe edildi. Araştırma 12 gün sürdü. Altı günlük adaptasyon fazını takip eden 6 günlük deneme fazında, RUSITEC sistemin 6 fermenteri 3 gruba ayrıldı. İlk iki fermentere hiçbir ilave yapılmayıp kontrol fermenterleri olarak kullanılırken, ikinci iki fermentere günde 250 mg biberiye ekstraktı, üçüncü iki fermentere ise yine günde 250 mg adaçayı ekstraktı ilave edildi. Araştırmanın deneme fazı boyunca her gün ruminal pH ve protozoon sayısı belirlenerek, amonyak azotu (NH3-N), uçucu yağ asitleri (UYA) ve yem kuru maddesi sindirilebilirliği analizleri için numuneler toplandı. Biberiye ve adaçayı ekstraktları ruminal pH, toplam UYA miktarı, propiyonat ve bütirat üretimi, toplam protozoon sayısı, NH3-N konsantrasyonu ve yem kuru madde sindirilebilirliğinde istatistiksel bir değişikliğe neden olmadı. Ancak, hiçbir ilavenin yapılmadığı kontrol fermenterleri ile karşılaştırıldığında biberiye ve adaçayı ekstraktları asetat üretimini azaltarak, asetatın propiyonata oranını düşürdü (P < 0,05). Sonuç olarak, bu araştırmadan elde edilen bulgular biberiye ve adaçayı ekstraktlarının ruminal fermantasyonunun bazı parametreleri üzerinde olumlu etkiler oluşturduğunu gösterdi. Bu maddelerin ruminant rasyonlarına ilavesi yine benzer etkiler meydana getirdiği takdirde hayvan verimliliğinde olumlu değişiklikler gözlenebilir.
