Histologic changes of adult rat testis in experimental hyperthyroidism

Aim: It was aimed to investigate histologic changes of adult rat testis in experimental hyperthyroidism. Materials and Methods: Thirty male Wistar albino rats, weighting 230-280 gr were used. Rats in group I (Control group) were given daily s.c. injections of 0.025 N NaOH in saline, rats in group II received daily s.c. injections of 250 mcg/kg L-tiroksin for 10 days and rats in Group III received daily s.c. injections of 250 mcg/kg L-tiroksin for 20 days. Thyroid function tests were quantified by Advia Centaur autoanalyzer. Histological analysis of testis tissue including Johnsen's testicular biopsy score was performed for each group after practicing routine histologic procedures. Results: Johnsen's testicular biopsy score of group II and group III were significantly decreased when com¬pared to group I (p<0.05). Although germ cell morphology of group II and group III were effected, interstitial cell morphology was not effected in hyperthyroidism. Conclusion: We have observed that spermatogenetic activity of adult rat testis was effected in experimental hyperthyroidism. This observation led us to conclude that it might be the result of direct effect of thyroid hor¬mones on germ cells and/or the result of indirect effect on germ cells via Leydig cells.

Amaç: Erişkin sıçanlarda deneysel hipertiroidi oluşturularak testis dokusundaki histolojik değişikliklerin ışık mikroskopik düzeyde incelenmesi amaçlandı. Materyal ve Metod: Ağırlıkları 230-280 gr olan 30 adet erişkin Wistar albino erkek sıçan kullanıldı. Grup I'e (Kontrol grubu) 10 gün serum fizyolojik, Grup Il'ye 10 gün ve Grup m'e 20 gün , günde bir kez 250 mcg/kg L-tiroksin subkutan enjekte edildi. Tiroid fonksiyon testleri, Advia Centaur otoanalizörde kemilüminesans yöntemi ile çalışıldı. Histolojik inceleme için rutin takip yöntemleri uygulandıktan sonra elde edilen testis dokusu kesitlerinde her grup için ayrı ayrı Johnsen testiküler biyopsi skoru hesaplandı ve histolojik değerlendirme yapıldı. Bulgular: Spermiyogenezin değerlendirildiği Johnsen testiküler biyopsi skoru açısından gruplar arasındaki fark istatistiksel olarak anlamlıydı (p<0.05). Grup İİ ve Grup İİİ'de germ hücre sayısının ve seminifer tübül histolojisinin değiştiği ancak interstisiyel alan¬da bulunan hücrelerin morfolojilerinin değişmediği saptandı. Sonuç: Erişkin dönemde oluşan hipertiroidinin, sıçan testisinde spermiyogenetik aktiviteyi etkilediği gözlendi. Bu değişikliğin, tiroid hormonunun doğrudan germ hücrelerine etkisi ve/veya Leydig hücreleri üzerinden dolaylı etkisi olabileceği sonucuna varıldı.
