Diarrheal illnesses are very common in communities and hospitals. In these cases, diagnostic approaches may be expensive and non cost-effective. Direct microscopic examination and stool culture are the methods commonly used for diagnosis of these cases. Detection of leukocytes in direct microscopic examination is very important in managing the treatment but in this method fresh stool samples must be used and the experience of the analyst is a major factor. Stool cultures require more complicated laboratory techniques. In this study, we explored stool lactoferrin levels to detect leukocytes as an inexpensive and simple method. For this purpose we exp¬lored anti-lactoferrin antibody coated latex particles agglutination in stool suspensions. In our study, we detected 93.75% (+), 68.75% (++) agglutination in specimens that were positive for leukocytes in direct microscopic examination. Sensitivity of our study was evalu¬ated to be 68.75%. In conclusion, we suggest that anti-lactoferrin antibody coated latex particle agglutination may be used to detect leukocytes in stool specimens.

Enteritler, hastanelerde ve toplumda oldukça sık görülen hastalıklar olduğundan, bu olgularda tanısal yaklaşımlar önemli bir sorun oluşturmaktadır. Bu konuda en sık uygulanan yöntem direkt dışkı mik-roskopisinde lökosit saptanmasıdır. Bu yöntem dışkının kısa süre içinde incelenmesini gerektirir ve yöntemi uygulayan kişinin bu konudaki yetkinliği de önemli bir faktördür. Dışkı kültürleri için ise gelişmiş laboratuvar koşulları gerekir. Biz bu çalışmada dışkıda lökositler ile laktoferrin arasındaki ilişkiyi araştırdık. Bu amaçla anti-laktoferrin antikoru kaplı lateks partikül-leri kullanarak dışkı süspansiyonlarında aglütinasyon arandı; lökosit belirlenen olguların %93.75'inde (+) ve %68.75'inde (++) olarak saptandı. Dışkı kültürlerinden sadece bir tanesinde üreme olması nedeniyle anlamlı bir karşılaştırma yapılamadı. Tüm olgular ele alındığında toplam %49.9 pozitif sonuç olması anlamlı olmadığından la-boratuvarımızda hazırlanan solüsyonla (++) olarak bulunan değerler temel alınarak çalışmamızın sensitivitesi %68.75 olarak değerlendirildi. Sonuç olarak antikor kaplı lateks partikülleriyle aglütinasyon saptanmasının dışkıda lökosit aranması için kullanılmasını öneriy¬oruz.
