Effect of aminoguanidine on renal polyamines and hydroxyproline levels in diabetic rats, Basement membrane (BM) thickening and expansion of mesangial matrix can be demonstra-ted in human kİdney within thc first two to three years after the onset of diabetes and wifhin the fırst fcw weeks of diabetes in the streptozotocİn (STZ) rat model. Certain growth factors are responsible for the BM thickening. Aminoguanidine (AG), an inhibitor of advanced glyca-tion end produets (AGEs) accumulation on tissue proteins, was demonstrated to prevent dia-betes-İnduced inerease of some growth factors. The polyamines, putrescine, spermidine and spermine are polycations that play an important role in celi growth and differentiation poliya-mines aı-e suggested to be involved in diabetic renal hypertrophy. In thİs study, experimental diabetes was produced by trealing male Wİstar albino rats with a single injection of STZ (50 mg/kg, i.v.) Diabetic rats are dıvided into two groups. One group (DM+AG) was given tap water containing aminoguanidin (lg/L), whereas the other group was kept as diabetic control (DM). After 10 weeks of treatment, animals were sacrificed by cardiac venİpuncture. Serum glucose levels were determİned enzymatically. Polyamine levels in kidney homogenate were measured by HPLC and hydroxyproline levels whİch are known as an index for tissue colla-gen were measured by spectrophotometrically İn glomerular basement membrane (GBM) pre-parations. Polyamine levels İn kİdney did not change in both diabetes and dİabetes+AG groups. But hydroxyproline levels in kidney inereased sİgnificantly in diabetes AG treatment ameliorated diabetes-induced changes in GBM hydroxyproline content. Some studies suggest that the enhancement İn polyamines is not a crucial determinant for kidney hypertrophy. Our results confirm these previous reports. Our experiments also demonstrate that renal hydroxy-proline levels inerease in diabetic rats and AG treatment prevents this inerement

Poliaminler (pııtresin, spermidin, spermin), hücre büyümesi ve farklılaşmasında önemli rol oynayan polikatyonlardır. Poliaminlerin dİabetik rcnal hipertrofinin ortaya çıkmasrnda rol oynadığı bildirilmiştir. Diabetin başlangıcından 2 ila 3 yıl sonra veya streptozotosin (STZ) ile dia-betİk yapılan sıçan modellerinde ilk birkaç haftada, böbrek mezangial matriksinde genişleme, bazal membranda kalınlaşma saptanmıştır. Büyüme faktörleri, glomerüler bazal mcmbran kalınlaşmasından büyük ölçüde sorumlu tutulmaktadır. İleri glikasyon ürünlerinin (AGE) oluşumunu inhibe eden Aminoguanİdin (AG)'in diabete bağlı büyüme faktörlerindekİ artışı önlediği ifade edilmiştir. Bu çalışmada, 50 mg/kg STZ enjeksiyonu ile deneysel diabet oluşturulan Wistar albino erkek sıçanlarda AG'in diabetik nefropati bulguları üzerine etkisi incelendi. Bu amaçla diabetik sıçanların bir grubuna lg/L dozda AG oral yolla 10 hafta süreyle verildi. Böbrek dokusu poliamİn düzeyleri HPLC yöntemi ile, hidroksiprolin düzeyleri spektrofotometrik olarak ölçüldü. Doku hidroksiprolin içeriği, kollajen indeksi olarak kullanıldı. Kontrol grubu sıçanlardaki putresin (16.0+3.7 nmol/g doku), spermidin (183.9+51.0) ve spermin (290.0+91.7) ortalama değerleri diğer iki diabetik grupta anlamlı bir farklılık göstermedi. Ancak böbrek hidroksiprolin düzeyleri, diabetik grupta anlamlı ölçüde aitti (kontrol: 1.95±0.61 pmol/g doku, DM: 3.43+1.05). AG uygulaması, diabete bağlı böbrek hidroksiprolin artışını engelledi (2.58±0.66).
