Apoptosis in polycystic kidney disease. Increased apoptosis is reported in different models of polycystic kidney disease. However, the role of apoptosis in cystogenesis and in fhe develop-ment of renal failure is not known. The Han:Sprague-Dawley (SPRD) rat model phenotypi-cally resembles human autosomal dominant polycystic kidney disease. Therefore, it presents a model to investigate the pathogenesis of this disease. Han:SPRD rats were studied during the early stages (1 day old and 2 weeks old) of their disease. Homozygous (Cy/Cy) rats had massive cystic development and renal failure at 2 weeks of age. Apoptotic cells were detected in kidney tissues by the TÜNEL (Tdt-mediated dUTP nick end-labeling) assay. TÜNEL (+) cells were significantly increased in 2-week-old heterozygous (Cy/+) and Cy/Cy rat kidneys compared to normal (+/+) littermate controls. TÜNEL (+) cells were observed both in cystic and non-cystic tubules. In conclusion, increased apoptosis may play a role in the development of renal failure in polycystic kidney disease.

Apoptoz artışı, çeşitli polikistik böbrek hastalığı modellerinde bildirilmiştir. Buna karşılık, apoptozun kist patogenezinde ve böbrek yetersizliği gelişimindeki rolü bilinmemektedir. Han:Sprague-Dawley (SPRD) sıçan modeli, insandaki otozomal dominant polikistik böbrek hastalığına fenotipik benzerlikler gösterir. Bu nedenle, bu hastalığın patogenezinin araştırılması için iyi bir modeldir. Han:SPRD sıçanları, hastalığın erken döneminde (1. gün ve 2. hafta) çalışmaya alındı. Homo-zigot (Cy/Cy) olan sıçanlarda 2. haftada yaygın kist oluşumu ile birlikte böbrek yetersizliği gelişimi gözlendi. Sonuç olarak, polikistik böbrek hastalığındaki apoptoz artışının böbrek yetersizliğinin gelişiminde rolü olabilir.
