Production of mıtlticellular tumor spheroids from tumor celi lines. The multicellular tumor spheroid model vvhich is of intermediate complexicity between in vivo tumors and monolayer cultures represents a system having a high correlation with tumors in vivo in cancer research. In this study we cultured spheroids using the MCF7, CC53İS and C6 tumor celi lines accor-ding to the liquid overlay technique. Although the general clıaracteristics of spheroid formati-on were similar in CC53İS and MCF7 celi lines, CC53İS spheroids showed a distinct pseu-doglandular structure under the light microscope. C6 spheroids displayed a continuous grovvtlı with large irregular aggregates of spheroid assembly. While profiles of apoptotic cells were detected in the inner regions of MCF7 and CC53İS spheroids, C6 spheroids had no necrotic centers. FM3A cells did not form spheroids. In conclusion, MCF7, CC53İS and C6 tumor cells foımed typical multicellular tumor spheroids which are suitable for use as a realistle model to study tumor response to antineoplastic agents and radiation.

Organizasyon bakımından tek tabakalı hücre kültürü ile in vivo tümör arasında yer alan multi-selüler tümör sferoid modeli in vivo tümör korelasyonu yüksek bir sistem olarak kanser araştırmalarında kullanılmaya başlanmıştır. Biz laboratuarımızda bulunan MCF7, CC53İS ve C6 tümör hücrelerinden sıvı üsttabaka yöntemine göre sferoidler oluşturduk. MCF7 ve CC53İS hücreleri genel sferoid oluşturma süreçleri açısından benzer özellikler göstermesine karşın CC53İS sferoidleri psödoglandüler yapılarıyla ışık mikroskobunda farklı bir görünüm sergilediler. C6 sferoidleri sürekli bir büyüme sürecinde birbirleriyle birleşerek büyük düzensiz kümelenmeler gösterdi. MCF7 ve CC53İS sferoidlerinin merkezi bölümlerinde apoptotik hücre profilleri saptanırken C6 sferoidlerinde nekrotik bölüm görülmedi. FM3A hücreleri sıvı üsttabaka yönteminde sferoid oluşturmadılar. Bu özellikleriyle MCF7, CC53İS ve C6 tümör hücrelerinin antineoplastik ilaç ve radyasyon uygulamaları için in vitro değerlendirmelerde kullanılmaya uygun sferoidler oluşturduğu saptandı.
