FLOW SİTOMETRE İLE NÖTROFİLLERİN OKSİDATİF PATLAMA AKTİVİTESİ ÖLÇÜLEBİLİR Mİ?

Can oxidative burst activity of neutrophils be measured byflow cytometry? Measurement of neutrophil functions are labor intensive and often very diffucult to standardize. In fhis study oxidative burst activity of neutrophils has been measured by modified techniques of flow cytometry. We activated the cells with phorbol 12-myristate 13-acetate. This induction causes an increase in oxidative burst activity which begins at the 5lh minute and reaches to %99 percent at 15* minute. The same method and Esclıerichia coli was used for phagocytosis activity. We found that 46% of cells showed phagocytosis activity at the 45* minute. According to these results, flovv cytometry can be used for measurement of oxidative burst activity and phagocytosis of neutrophils.

Nötrofil fonksiyonlarının ölçüm yöntemleri oldukça zahmetlidir ve standardize edilmeleri zordur. Nötrofil fonksiyonları flow sitometre yöntemleri ile incelenebilmektedir. Bu çalışmada nötrofillerin oksidatif patlama aktivitesi flow sitometre'de modifiye edilmiş tekniklerle ölçüldü. Hücreler oksidatif patlama için phorbol 12-myristate 13-acetate ile uyarıldı. Çalışmada nötrofillerin spontan oksidatif patlama aktivitesi çok düşük olmasına rağmen, uyarıldıktan sonra 5. dakikada başlayarak ve 15. dakikada %99'a ulaşmaktaydı. Fagositoz aktivitesi için aynı metot ve Esclıerichia coli kullanıldı. Hücrelerin %46'sı 45. dakikada fagositoz yapmaktaydı. Bu çalışmadaki bulgularımıza göre flow sitometre, oksidatif patlama ve fagositoz gibi nötrofil fonksiyonlarının ölçümünde kullanılabilecek bir metotdur.
