Many studies have reported that melatonin has a relationship with kidney functions directly or indirectly. The aim of this study was to examine the possible histological changes at renin granules in the juxtaglomerular apparatus in the kidneys after chronically injection of melatonin. Twenty six female Wistar rats were divided into two groups: control (C) group (n=11) and chronically melatonin injected (M) group (n=15). Melatonin was injected subcutaneously 1 mg/kg daily in a volume of 2 cc for 3 weeks. At the end of the experiment, serial sections of kidneys were stained with Hematoxylen-eosin, periodic acid schiff (PAS) and Van Gieson stain. In Hematoxylen-eosin and Van Gieson stained kidneys, no different histological changes was observed in both group. In the PAS stained kidney sections, an increase has been detected in the renin granules (juxtaglomerular cells), qualitatively in the M group. It is thought that this increase might be the regulatory effect of melatonin on kidneys and blood pressure.

Birçok çalışmada melatoninin böbrek fonksiyonları ile doğrudan veya dolaylı ilişkisi olduğu bildirilmiştir. Bu çalışmanın amacı kronik melatonin enjeksiyonundan sonra böbrekteki jukstaglomerüler apparatusta bulunan renin granüllerindeki olası histolojik değişiklikleri araştırmaktı. Yirmi altı adet Wistar cinsi dişi sıçan iki gruba bölünmüştür: kontrol (K) grubu (n=ll) ve kronik olarak melatonin enjekte edilen (M) grup (n=15). Melatonin 3 hafta boyunca hergün, subkutan olarak l mg/kg dozunda ve 2 cc hacimde enjekte edildi. Deneyin sonunda böbrekten alınan seri kesitler Hematoksilen-eosin, periodik asit schiff (PAS) ve Van Gieson boyaları ile boyandı. Hematoksilen-eosin ve Van Gieson ile boyanmış böbreklerde iki grup arasında herhangi bir farklılık gözlenmedi. PAS ile boyanan böbrek kesitlerinde ise M grubunda renin granüllerinde (jukstaglomerüler hücrelerde) niteliksel olarak bir artış gözlendi. Bu artışın melatoninin böbrekler ve kan basıncı üzerindeki düzenleyici etkisi sonucu oluştuğu düşünüldü.
