Histological Effects of Zinc or Melatonin Deficiency on Testes of the Rats

The present study was designed to investigate the histological effects of zinc and/or melatonin deficiency on rat testes. A total of 24 adult male Spraque Dawley rats were used in the study. The experiments were done in Selçuk University Experimental Medicine Research and Application Center. The rats were divided into 4 groups. Group I got zinc deficient diet (n=6). Pinealectomy was applied to the Group II (n=6). Group HI consisted of pinealectomized (Px) and zinc deficient rats (n=6). Group IV members were intact control rats (n~6).
At the end of the 3 weeks all animals were decapitated and plasma zinc and melatonin levels were determined. Testicular tissues were stained by hematoxilene eosine and investigated under the light microscope. The seminiferous tubules and spermatogenetic activation was observed and spermatogenesis was scored.
The plasma zinc levels in Px group (Group II) were significantly lower than those of control (p<0.01). In the Group I and Group III plasma zinc levels were significantly lower than those of not only control but also Group II (p<0.01 for all). The melatonin levels in Group I, II and III were significantly lower than control (p<0.01). In spermatogenetic evaluation, there was no significant difference between control and Group II. A significant suppression was observed in the spermatogenetic activity of the zinc deficient groups (Group I and Group III, p<0.01). The suppression was higher in the Group I than Group HI
In the seminifer tubules of the Group I, degeneration, necrosis, obstruction and some hyalinisation were observed. In Group 111 an increase in the basal membrane thickness, lymphocytic infiltration and necrosis in some germ cells were observed. No significant histological changes were obtained in the Group II compared with the control.
The findings of this study support the previous studies that zinc deficiency causes testicular damage. These results also suggest that melatonin deficiency may reduce the testicular damage caused by zinc deficiency.

Bu çalışmada melatonin ve çinko eksikliğinin birlikte ve ayrı ayrı rat testisleri üzerindeki histolojik etkileri araştırıldı. Çalışma Selçuk Üniversitesi Deneysel Tıp Araştırma ve Uygulama Merkezinde yapıldı. Yirmi dört adet yetişkin erkek Spraque Dawley rat dört gruba ayrıldı. I. gruba çinko eksik diyet verildi (n=6). II. gruba pinealektomi uygulandı (n-6). III. gruba hem pinealektomi uygulandı, hem de çinko eksik diyet verildi (n=6). IV. grup kontrol olarak kullanıldı (n=6).

haftanın sonunda hayvanlardan dekapitasyonla alınan kan örneklerinde plazma çinko ve melatonin düzeyleri tayin edildi. Testis doku kesitleri hematoksilen eozin ile boyandı ve ışık mikroskobunda incelendi. Spermatogenez skorlaması ile spermatogenetik faaliyetler değerlendirildi.
II. grupta plazma çinko seviyeleri kontrol grubundan anlamlı olarak daha düşüktü (p<0.0l). I. ve III. Grupta plazma çinko seviyeleri hem kontrol, hem de II. gruptan anlamlı şekilde daha düşüktü (p<0.01). Grup I, II ve III'deki melatonin seviyeleri kontrol grubuna göre daha düşük bulundu (p<0.01). Pinealektomize grupların (Grup II ve III) melatonin düzeyleri grup I'den de anlamlı derecede düşüktü (p<0.01). Spermatogenetik değerlendirmede grup II ile kontrol grubu arasında önemli bir fark görülmedi. Grup I ve İH'de spermatogenetik aktivitede bir baskılanma gözlendi (p<0.01). Grup I'deki baskılanma grup IH'deki baskılanmadan daha yüksekti. Grup I'de tübüler dejenerasyon ve nekroz, tübül lümenlerinde tıkanma, bazı tübüllerde hiyalinizasyon gözlendi. III. grupta bazal membran kalınlaşması, lenfositik infiltrasyon ve bazı germ hücrelerinde nekroz gözlendi. Grup II ve kontrol grubunda histopatolojik değişiklik belirlenmedi.
Bu çalışmanın bulguları literatür bilgilerine uygun olarak çinko eksikliğinin testiküler harabiyete yol açtığını göstermektedir. Melatonin eksikliği, 3 haftalık çinko eksikliğinin yol açtığı testiküler hasarı azaltabilir.
