Culture studies of the esophageal tissue have mostly been on the investigation of mucosal changes. In this study, we aimed to form functional esophageal segment in vitro and to calculate and compare the growth kinetics of esophageal muscle cells of fetal and adult distal esophagus. The muscular coat of distal 1/3 esophageal segments of adult and fetal New Zealand rabbits were cultured and identified as smooth muscle cells (SMC) by immunostaining for anti-actin and anti-myosin antibodies. Growth kinetics of SMCs were calculated and compared using population doubling time (PDT) and thymidine labeling index (TLI). Viability rates of all cells were calculated using trypan blue. The ratio of cell populations stained with anti-actin and anti-myosin antibodies was higher than 90% and the cells were characteristic spindle shaped, same as the muscle cell morphology. Calculated PDT of fetal and adult SMCs were 145,5 and 95,2 hours respectively and TLI values were found to be similar. Fine dissection of the tissue and appropriate medium is mandatory to achive isolated esophageal SMCs. The fetal esophageal cells showed proliferation slower than the adults. Conclusion, we can spaculate that smooth muscle cells obtained from different tissues act similar in the same culture condition.

Özofagus doku kültür çalışmaları çoğunlukla mukozal değişiklikleri incelemeye yöneliktir. Çalışmamızda, tavşan modelinde in vitro fonksiyonel özofagus segmenti hazırlamaya yönelik olarak, fetus ve erişkin distal özofagus düz kas hücrelerinin morfolojik özellikleri ve büyüme kinetiklerinin hesaplanarak karşılaştırılması amaçlandı. Erişkin ve fetus Yeni Zelanda tipi tavşanların özofaguslarının distal 1/3 kısımındaki dokuların kas tabakası kültüre edildi ve üreyen hücreler anti-aktin ve anti-miyozin antikorları ile immünositokimyasal boyama yapılarak tiplendirildi. Popülasyon kinetikleri, popülasyon sayısını ikiye katlama zamanı (Population doubling time, PDT) ve timidinle işaretlenme indeksi (Thymidine labelling index, TLI) kullanarak hesaplandı ve karşılaştırıldı. Trypan blue ile boyanarak hücrelerin canlılık oranları hesaplandı. Heriki hücre popülasyonu, anti-aktin ve anti-miyozin antikorları ile % 90’ın üzerinde pozitif boyandı ve hücrelerin iğ şeklinde, düz kas morfolojisine sahip oldukları görüldü. Fetal ve erişkin hücrelerin PDT değerleri sırasıyla 95,2 ve 145,5 saat, TLI değerleri ise birbirlerine yakın bulundu. Uygun besiyeri ve yeterli doku disseksiyonu ile kolaylıkla izole özofagus düz kas hücre kültürü elde edilebilmektedir. Erişkin özofagus düz kas hücrelerinin, kültür ortamında daha hızla çoğaldıkları gözlendi. Sonuç olarak, farklı dokulardan elde edilen düz kas hücrelerinin kültür ortamında benzer üreme özellikleri gösterdikleri söylenebilir.
