This study aimed to assess the effect of L-Arginine, a nitric oxide (NO - ) procursor, and CaEDTA, an extracellular zinc chelator, on the short time epileptic seizure which was formed by pilocarpine HCl injection. The experiment was consist of 4 groups, including seizure control (saline 10 l, i.c.v., and then 380 mg/kg pilocarpine HCl i.p.), L-Arginine (150 g/10 l, i.c.v.), CaEDTA (100 mM, 10 l, i.c.v.), L-Arginine+CaEDTA groups. Seizure behaviour of the rats were observed for two hours following the injections and the animals were sacrificed at the end of the process and the brains were taken out for immersion Autometallography (iZnS AMG , NeoTimm) and N-6- methoxy-8-quinolyl-para-toluenesulfonamide (TSQ) staining processes. Seizure behaviour in the groups of seizure control, L-Arginine, CaEDTA and L-Arginine+CaEDTA was found to be strong and equal to each other. Moreover, in these groups no zinc translocation was spotted in hippocampal stainings which were performed by using zinc histochemistry. With these results, we concluded that the NO - procursor injection alone, zinc chelation alone or together had no effects especially on seizure activity and zinc depletion in the hippocampus during and at the end of the two hour epileptic seizure.

Bu çalışmada, bir nitrik oksit (NO - ) prekürsörü olan L-Arginine ve bir ekstrasellüler çinko şelatörü olan CaEDTA’nın pilokarpine HCl ile oluşturulan kısa süreli epileptik nöbet üzerine etkileri araştırıldı. Deney, nöbet kontrol (serum fizyolojik 10 l, i.c.v., ve sonra 380 mg/kg pilokarpine HCl i.p.), L-Arginine (150 g/10 l, i.c.v.), CaEDTA (100 mM, 10 l, i.c.v.), L-Arginine+CaEDTA olmak üzere 4 gruptan oluştu. Enjeksiyonlardan sonra iki saat boyunca nöbet davranışları gözlemlenen sıçanlar bu süre sonunda sakrifiye edilerek immersion Autometallography (iZnS AMG , NeoTimm) ve N-6-methoxy-8-quinolyl-para-toluenesulfonamide (TSQ) boyamaları için beyinleri çıkarıldı. Nöbet kontrol, L-Arginine, CaEDTA ve L-Arginine+CaEDTA gruplarında nöbet davranışları şiddetli ve birbirleri ile eşit düzeyde bulundu. Bu gruplarda çinko histokimyası için yapılan hipokampal boyamalarda çinko translokasyonuna rastlanmadı. Bu bulgular bizi iki saatlik epileptik nöbet sırasında ve sonunda NO - prekürsörü enjeksiyonunun, çinko şelasyonunun veya her ikisinin özellikle nöbet aktivitesine ve hipokampusda çinko deplesyonuna etkili olmadığı sonucuna götürdü.
