The aim of study was to determine the effect of 4,6 dinitro-o-cresol (DNOC) on glucose 6-phosphate dehydrogenase (G6PD) and malate dehydrogenase (MDH) activities in some tissues of rats. While glucose G6PD is a control enzyme in pentose phosphate pathway, MDH is an important enzyme in many pathways. In our study, rats (Rattus norvegicus) were intraperitoneally injected with 2.8 mg kg -1 dose of DNOC, a toxic agent widely used for insect control and crop protection. At 0, 2, 4, 8, 16, 32, 64 and 72 hours following injection, the liver, kidney, brain and small intestine tissues of rats were used to determine G6PD activity; on the other hand only liver and kidney tissues were used to determine MDH activity. Enzyme activities were measured spectrophotometrically. DNOC did not cause important changes in the G6PD activity in both liver and kidney tissues (p>0.05). It caused significant changes in brain and small intestine. In addition, it led to a decrease in MDH activity in both liver and kidney in various hours (p<0.05).

Çalışmanın amacı sıçanların bazı dokularında malat dehidrogenaz (MDH) ve glukoz 6-fosfat dehidrogenaz (G6PD) aktiviteleri üzerine 4,6 dinitro-o-krezol’ün (DNOC) etkisini araştırmaktır. G6PD pentoz fosfat metabolik yolunun kontrol enzimiyken, MDH birçok metabolik yolun önemli enzimidir. Çalışmamızda, yaygın bir şekilde böcek kontrolünde ve ürün korunmasında kullanılan ve toksik bir ajan olan DNOC’un 2.8 mg kg -1 ’lık dozu, sıçanlara (Rattus norvegicus) intraperitonal yolla enjekte edildi. Enjeksiyondan 0, 2, 4, 8, 16, 32, 64 ve 72 saat sonra G6PD aktivitesi için sıçanların karaciğer, böbrek, beyin ve incebağırsak dokuları, MDH aktivitesi için ise sadece karaciğer ve böbrek dokuları deneye alındı. Enzim aktiviteleri spektrofotometrik yolla ölçüldü. DNOC, hem karaciğer hem de böbrek dokusunda G6PD aktivitesinde önemli bir değişiklik meydana getirmemiştir (p>0.05). Beyin ve incebağırsak dokusunda ise anlamlı değişikliklere neden olmuştur. İlave olarak, MDH aktivitesinde hem karaciğerde hem de böbrek dokusunda bazı saatlerde azalmalara sebep olmuştur (p<0.05).
