The effects of ethanol on devetopment and morphogenesis of the mammalian embryos at the stage of early organo-genesisr and the role offree radicals in this effect.

Aim: The excessive maternal alcohol drinking results in Fetal Alcohol Syndrome which is a pathoiogy caused by the embryotoxlc effects of ethanol in prenatal period. The mechanisms of ethanol action at the ccliular or molecular Ic-veis are scarce. İn this study it was aimed to investigate the effects of ethanol on grovvth and development of mam¬malian embryos as well as the morphological strueture, and the role offree radicals on these effects. Meîhods: Wistar rats vvere obtained from the Selçuk University Experimental Medlcine Research Center and thelr 9.5 days embryos were explanted and cultured for 48 hours in rat serum. VVhole rat serum was used as a culture medium for the cont-rol group whiie different concentrations of ethanol (250-500 mg%) vvere added to rat serum for the experimental groups. Also, the lowest effective concentration of ethanol for ali parameters was added to the culture media in the presence of an antloxidant superoxide dismutase (SOD). Ten embryos vvere used for each experimentai condıtıon Dose-dependent effects of ethanol on embryonic developmental parameters such as; total morphological score. yolk sac diameter, crown-rump length, somite number, embryo and yolk sac protein contents vvere compared using morp¬hological and biochemical methods. Each embryo vvas evaluated for the presence of any malformations. Results: Compared to the control embryos, ethanol signifıcantly decreased ali developmental parameters dose-dependently (p<0.05} vvith an inerease in overall dismorphology. The haematoma in different regions, especlally in the neural tube was most freguent/y observed. vVhen the SOD was added to the culture media in the presence of ethanol, grovvth and developmental parameters vvere improved (p<0.05) and there vvas a decrease in the incidence of malformations (p<0.05). Conclusion: A dose dependant developmental toxicity of ethanol on rat embryos during organogenesls vvas determlned, these effects ınight involve free oxygen radicals.

Amaç: Gebe kadınların aşırı alkol almaları Fötal Alkol Sendromu adı verilen ve etanolun prenatal dönemde embriyo-toksik etkileri sonucu ortaya çıkan bir patolojiye neden olmaktadır. Etanolun bu dönemde hücresel ve moleküler düzeydeki etki mekanizmaları ise tam olarak bilinmemektedir. Bu çalışmada etanolun embriyonik büyüme ve gelişme ile morfolojik yapı üzerine olan etkileri ve serbest radikallerin bu etkilerdeki rolünün araştırılması amaçlandı. Yöntem: Selçuk Üniversitesi Deneysel Tıp Araştırma ve Uygulama Merkezi hayvan laboratuvarından elde edilen Wistar ratlardan gebeliklerinin 9.5 uncu gününde diseke edilerek çıkartılan embriyoların rat serumu içerisinde 48 saat süreyle kültürü yapıldı. Kültür ortamı olarak: kontrol grubu için normal rat serumu kullanılırken, deney grupları için rat serumuna değişen konsantrasyonlarda etanol (250-500 mg%j ilave edildi. Ayrıca etanolun toksik etkisinin tüm parametreleri etkile¬diği dozu ile birlikte antloksldan superoksld dısmutaz (SODJ kültür ortamına ilave edildi. Her bir konsantrasyon için 10 rat embriyosu kullanıldı. Etanolun rat embriyolarının gelişim parametreleri (totai morfolojik skor, yolksaik çapı, tepe-kıç mesafesi, somit sayısı, embriyo ve yolk saik protein içerikleri) üzerine doz bağımlı etkileri, morfolojik ve biyokimyasal yöntemlerle karşılaştırıldı. Embriyolar maiformasyon varlığı açısından da değerlendirildi. Bulgular: Kontrol embriyoları ile karşılaştırıldığında etanol'ün doz bağımlı olarak bütün gelişimsel parametreleri gerilettiği (P<0.05} ve genel mor¬folojide bozukluklara sebep olduğu gözlendi. Özellikle nöral tüp olmak üzere değişik bölgelerde hematomlar gözlen¬di. Kültür ortamına etanol ile birlikte SOD eklendiğinde büyüme ve gelişme parametrelerinde artış (P<0.05} ve maifor¬masyon sıklığında azalma (P<0.05} tespit edildi. Sonuç: Etanol'ün organogenez dönemindeki rat embriyoları üzerine doz bağımlı gelişimsel toksisiteye sebep olduğu ve bu etkilerde serbest oksijen radikallerinin rol oynayabileceği belirlendi.
