In this study, the effect of benzene which is known as a carcinogen and toxic agent on liver
Glutathione S-transferase (GST) enzyme was studied in vitro. GST enzyme activity was determined by
monitoring the formation of 1-chloro-2,4-dinitrobenzene -Glutathione (CDNB-GSH) conjugate by
spectroscopic method.
Our results showed that there is an inverse correlation between GST conjugation activity and
concentration of benzene, as well as reaction time. Variations in the Vmax and Km value for CDNB ofthe enzyme was observed. The results obtained showed that GST activity for CDNB-GSH conjugation reaction was inhibited with mixed reaction mechanism by benzene.

Bu çalışmada, karsinojen ve toksik bir ajan olarak bilinen benzenin karaciğer Glutatyon Stransferaz
(GST) enzimi üzerine etkisi in vitro çalışıldı. GST enzim aktivitesi, 1-kloro-2,4-dinitrobenzenglutatyon
(CDNB-GSH) konjugatının oluşumu spektroskopik izlenerek tayin edildi.
Sonuçlarımız, GST konjugasyon aktivitesi ile benzen derişimi ve tepkime zamanı arasında ters
bağlantı olduğunu gösterdi. Enzimin CDNB için Vmax ve Km değerlerinde farklılıklar gözlendi. Elde
edilen sonuçlar, GST enziminin CDNB-GSH konjugasyon tepkimesini benzenin karışık inhibisyon
mekanizması ile inhibe ettiğini gösterdi.
