HLA DQA1 loci are highly informative polymorphic loci that are gaining popularity for identity testing. HLA DQA1 system easy to work with, fast and reliable. This loci with PCR technique is a very promising tool for genetic investigations in both paternity and crime cases. Design.- Human genomic DNA was extracted by Chelex(r) 100 procedure. The amplification of HLA DQA1 locus was performed by single locus PCR reaction in the case of HLA DQA1 locus according to the manufacturer's recommendations using the Perkin Elmer AmpliType DQA1 PCR Amplification Typing Kits. The electrophoresis of PCR products of HLA DQA1 are carried out on agarose gel. Results.- It was shown from the obtained bands that the DNA is amplified. Hybridization is made from the PCR products by Perkin Elmer AmpliType DQA1 PCR Amplification Typing Kits and that typed as 2,3 as a result and this allele was compared with the Capillary electrophoreograms. Conclusion.- In this study, Capillary gel electrophoresis (CGE) was performed on PCR products of HLA DQA1 due to its advantages are ultra-resolution, ultralow sample volume, extremely high efficiency, rapid separation time, easy quantitation and amenability to automation.

HLA DQA1 lokusu Adli bilimler araştırmaları için hızlı, kolay, güvenilir bilgi alınabilen polimorfik DNA lokusudur. Bu lokus, PCR sayesinde nesep tayini ve Adli olayların aydınlatılmasında son derece umut verici bilgiler sunar. İnsan kaynaklı genomik DNA kelatlaştırma (Chelex(r) 100 ) yöntemiyle izole edildi. HLA DQA1 lokusunun çoğaltılması, Perkin Elmer AmpliType DQA1 PCR amplifikasyon ve tipleme kitinin kullanma kılavuzuna uygun olarak yapıldı. HLA DQA1 in PCR ürünlerine agaroz jel elektroforezi uygulandı. Elde edilen elektroforez bantlarından DNA nın çoğaldığı tespit edildi. Çoğalan PCR ürünlerinin Perkin Elmer AmpliType DQA1 PCR amplifikasyon ve tipleme kitiyle hibridizasyonu yapıldı ve 2,3 olarak tiplendirildi. Bu alel kapiller elektroforeogramları ile karşılaştırıldı. Bu çalışmada, kapiller jel elektroforez (CGE)'nin az miktarda örneğe gereksinim duyması, hızlı ve çabuk ayırım süresi, yüksek ayırıcılığı, kolay miktar hesabı ile otomasyona uygunluğu gibi özelliklerinden yola çıkılarak HLA DQA1 in PCR ürünlerinin ayırımı amaçlandı.
