Genome related studies have been progressed slowly in most plants due to their large genome size and complex genome structure. Therefore it is becoming important to evaluate applicability of results belonging to certain species for other species that can be genetically linked. In this study, DNA from a bread wheat variety ‘Gerek-79’, growing in wide area of Turkey, was amplified with 29 barley microsatellite primers by PCR. Allele sizes of radioactively labeled PCR products were compared with those obtained with same primers in barley. Based on allele sizes, microsatellites producing products at similar length for wheat were determined as HVM15, HVM20, HVM23, HVM26, HVM31, HVM36, HVM40, HVM44, HVM54 and HVM62. In future studies, level of polymorphism among different wheat varieties will be determined based on those microsatellites from this study and possibility of using them for genotyping studies being addressed.

Genomik çalışmalar çoğu bitkinin oldukça büyük genoma sahip olmaları ve kompleks genom yapısı göstermeleri nedeniyle yavaş ilerleme göstermektedir. Bu nedenle bir bitki türünden elde edilen araştırma sonuçlarının o tür ile genetik ilişkisi olabilecek bir başka tür üzerinde uygulanabilirliğinin araştırılması önem kazanmaktadır. Bu araştırmada, Türkiye’de fazla miktarda yetiştirilen bir ekmeklik buğday çeşidi olan ‘Gerek-79’’un DNA’sı 29 arpa mikrosatelitinin her birine özgü polimeraz zincir reaksiyonu (PCR) primerleri kullanılarak çoğaltılmıştır. Buğdaydan elde edilen radyoaktif PCR ürünleri, daha önce arpada bu primerlerin kullanılması sonucu elde edilmesi muhtemel ürünlerin alel uzunlukları ile karşılaştırılmıştır. Alel uzunluklarına göre buğdayda benzer uzunlukta amplifikasyon ürünleri oluşturan mikrosatelitler HVM15, HVM20, HVM23, HVM26, HVM31, HVM36, HVM40, HVM44, HVM54 ve HVM62 olarak saptanmıştır. Bundan sonraki çalışmalarda belirlenen mikrosatelitlerin farklı buğday çeşitlerindeki polimorfizm özellikleri saptanarak, buğday genotiplendirme çalışmalarında kullanılma olasılığı incelenecektir.
